| 摘 要： | 本文合成毒死蜱的半抗原并对其进行结构鉴定，将半抗原与牛血清白蛋白和鸡卵清蛋白共价偶联分别制备免疫原和包被原，再取已免疫的Balb/c小鼠脾细胞与SP2/0小鼠骨髓瘤细胞融合，建立2株分泌毒死蜱单克隆抗体的杂交瘤细胞。以琼脂双扩散法鉴定抗体亚类类型，间接竞争ELISA方法测定了抗体对毒死蜱的灵敏度、特异性和亲和性。结果表明该抗体分泌IgG1亚类的单克隆抗体，且与毒死蜱的亲和性较高(1.5×107 L / mol)，与其他结构类似物的交叉反应均小于1%。本研究成功制备了毒死蜱特异性的单克隆抗体，为今后ELISA方法的建立提供了条件。

| Abstract: | Hapten of chlorpyrifos was synthesized and the structure was identified.The hapten was covalently conjugated to BSA and OVA to prepare immunogen and coating antigen.Two hybridoma cell lines secreting monoclonal antibodies(McAb)against chlorpyrifos had been established by fusing mouse myeloma cells SP2/0 and splenocytes from the Balb/c mouse immunized with immunogen.The immunoglobulin class was analyzed.The affinity and the specificity of McAb were determinated with indirect enzyme linked immunosorbent assay... |
