| 摘 要： | 本研究通过PCR-RFLP建立了直接利用基因组DNA检测尿苷酸合酶缺乏症（DUMPS）突变的方法.对济南市6个奶牛场224头奶牛及53头荷斯坦种公牛进行了DUMPS的PCR-RFLP检测,共检出2头杂合母牛（携带者）,杂合子占检测母牛群的0.89%,在荷斯坦公牛中只检测到一种基因型,没有发现隐性突变基因的携带者.实验证明,该方法重复性好、灵敏性高、稳定性可靠.

| Abstract: | Deficiency of uridine monophosphate synthase(DUMPS) is a monogenic autosomal recessive disorder in cattle,resulting in early embryonic death of homozygous offspring.To identify the mutation responsible for DUMPS,a direct genome DNA test based on PCR-RLFP was developed.Altogether 224 dairy cows from 6 herds in Jinan and 53 Holstein bulls were analysed,of which 2 cows,i.e.0.89% were confirmed DUMPS carriers in cows,and zreo in bulls.Experimention tested the method was repeatable,sensitive and stable. |
