产Iturins族脂肽生防芽孢杆菌的分离、筛选及鉴定

摘    要:为获得能够产Iturins类脂肽的芽孢杆菌以应用于植物的生物防治中,以果蔬体内、根际土壤等多生境中分离的118株细菌为出发菌株,采用平板对峙法、管碟法对其进行了初筛和复筛。结果表明:其中3株菌株对立枯丝核菌、瓜果腐霉和尖孢镰刀菌显示出强烈的抑制作用;通过薄层层析等方法对产Iturins脂肽菌株进行初步分析,最终确定K103为目标菌株。通过形态学鉴定、生理生化试验及16SrDNA的分子生物学等方法,对产抗菌脂肽的K103菌株进行分类学鉴定,最终确定菌株K103为解淀粉芽孢杆菌,并将其命名为解淀粉芽孢杆菌K103(Bacillus amyloliquefaciens K103)。

Abstract:In order to obtain Bacillus sp.strains producing Iturins for applications in biological control,118 bacterial strains from some vegetable or rhizosphere soil were used in preliminary and secondary screen by the plate confrontation tests and cylinder-plate method. Among them 3 strains,showing strong antifungal activity against Rhizoctonia solani,Fusarium oxysporum and Pythium aphanidermatum,were analyzed by thin layer chromatography and other methods to detect the ability of the strains to produce Iturins.Finally,the strain K103 was assigned as the target bacterial strain.The organism of interest was identified as a Bacillus amyloliquefaciens strain,and nominated as B.amyloliquefaciens K103 using biochemical,physiological and 16S ribosomal DNA sequencing.
