摘    要:应用ISSR标记,对广东地区30份野生狗牙根进行遗传多样性研究。从50个ISSR引物中筛选出15个多态性明显、反应稳定的引物,共扩增出151条谱带,平均每个引物能扩增出10.07条带,其中多态性条带总数为142条,多态性条带比率达93.7%。材料间遗传相似系数GS=0.58278~0.92715。基于遗传相似系数,利用UPGMA聚类分析表明,供试材料可聚为4类。POPGENE分析软件结果表明,平均Shannon信息指数I=0.5689,平均Nei's基因多样性h=0.3813,每位点平均有效等位基因数ne=1.6196。

Abstract:Thirty wild bermudagrass resources[Cynodon dactylon(Linnaeus) Persoon] in Guangdong were studied for an analysis of genetic diversity with ISSR. Fifteen effective primers selected from 50 ISSR primers were used for PCR, and 151 DNA fragments were amplified from 30 samples to give 142 fragments that were polymorphic, an average of 10.07 DNA bands per primer, the average percentage of polymorphic bands was 93.7%. The genetic similarity among all accessions ranged from 0.58278 to 0.92715, indicating a comparatively rich genetic diversity among the tested accessions. The accessions were divided into four groups based on UPGMA cluster genetic similarity. POPGENE analysis indicated that the average information index(I) of Shannon was 0.5689, gene diversity(h) of Nei was 0.3813, and effective number of alleles (ne) was 1.6196.
