不同株鸡新城疫病毒结构蛋白的比较研究

对鸡新城疫病毒(NDV)B1株、La Sota株、Mukteswar株和长春强毒野毒株(C87E7)感染的鸡胚尿囊液分别进行浓缩和提纯,并作SDS-PAGE和Western印迹,进行结构蛋白及其抗原性的分析。结果,4株NDV的电泳图谱显示11-12条结构蛋白带,分子量从43000到120000不等。其中各株均有3条主要蛋白带,8～9条次要蛋白带。不同株NDV的次要蛋白带有明显的区别。Schiff氏试剂染色证明,分子量为76000、52000和50000的蛋白为糖蛋白。结构蛋白的抗原性分析表明,4株NDV蛋白带中有2条具有共同抗原性,其他蛋白带的抗原性则有差异或明显不同。

A Comparative Study on Structural Polypeptides of Some Newcastle Disease Virus Strains

Some strains of Newcastle disease virus were concentrated and purified from allantoic fluid of chicken embryoes infected with B1, La Sota, Mukteswar and Changchun wild strain(C87E7). Virus structural proteins were run on sodium dodecylsulphate(SDS) -polyacrylamide gel discontinuous system and assayed for antipenicity with Western blotting by strain specific antisera prepared by immunizing chickens with respective virus strains. Electrophoretogram showed 11 to 12 structural protein bands with Mr from 43000 to 120000. Stained with Schiff's reagent, two glycoproteins with Mr 76000 and 52000 or 50000 were revealed of 11 or 12 structural proteins, 3 are major proteins and the other 8 or 9 are minor. There are significant differences in some minor proteins from different strains. And of all the immunoenzymatically examined bands of each of the 4 strains, two bands are common in antigenicity , others different significantly.
