绵羊体外受精技术条件优化的研究

摘    要:探讨了卵泡液、受精时间和季节对绵羊卵母细胞体外培养效果的影响。从屠宰场采集羊卵巢,抽取卵巢表面2mm-6mm的卵泡卵母细胞,进行体外成熟培养;卵母细胞成熟后,与处理后的精子共同培养进行受精。结果表明,在卵母细胞成熟培养液中添加绵羊卵泡液(SFF)组的卵裂率和囊胚发育率均优于添加同等浓度的牛卵泡液(BFF)组;受精22、18、12h组的卵裂率分别为64.85%、54.39%、48.72%,受精22h和18h组间差异显著(P〈0.05),受精22h组和12h组间差异极显著(P〈0.01)。秋季绵羊体外受精的卵裂率和囊胚率均高于夏季,差异显著(P〈0.05),但秋季与春季相比差异不显著(P〉0.05)。因此,进行绵羊卵母细胞体外成熟培养时,培养液中添加SFF;受精时,精卵共培养时间为22h的效果较好。在绵羊的繁殖季节秋季进行体外受精较春季和夏季的结果好。

Abstract:The effects of follicular fluid, seasons, fertilization time on sheep oocyte in vitro fertilization were investigated in the present experiment. Ovaries were collected at a local abattoir, and oocytes obtained from 2 mm-6 mm follicle were cultured. Matured oocytes were co-cultured with treated spermatozoa. The results showed that cleavage rate, blastocyst rate of SFF group in which SFF were added to oocyte maturation media were better than that of BFF group. The cleavage rate of fertilization for 22 h , 18 h and 12 h were 64.85%, 54.39%and 48.72%,respectively, there was significant difference between fertilization for 22 h to 18 h(P<0.05), and there was extremely significant difference between fertilization for 22 h to 12 h(P<0.01). Cleavage rate and blastocyst rate in autumn were higher than that in summer, and there was significant difference between autumn and summer(P<0.05), but there was no significant difference between autumn and spring (P>0.05). In conclusion, when SFF was added to oocytes maturation media, and sperm-oocyte co-culture for 22 h, the results were better than that in other groups in sheep IVF, and the results in autumn season were better than that in spring and summer.
