耐热白桦茎叶高效再生体系的建立

摘    要:耐热白桦是白桦的一个耐热型品种,在华北地区生长良好。本试验对其茎段、叶片外植体消毒,分化培养基、生根培养基筛选等进行了研究,筛选出耐热白桦的外植体消毒以浸泡75%酒精溶液中30s,2%次氯酸钠溶液浸泡10min为宜;叶片最佳分化培养基为:WPM+0.5 mg/L 6-BA+0.1 mg/L IBA,分化效率达94%,茎段最佳分化培养基为:MS+1 mg/L 6-BA+0.1 mg/L IBA,分化效率达92%;生根培养基初代培养时最佳为1/2MS+0.2 mg/L IBA,生根率达96.7%,继代培养时最佳为wpm+0.3 mg/L IBA,生根率达100%。本试验建立了耐热白桦高效再生体系,对其种苗高效繁殖实现园林推广应用具有重要意义。

Abstract:Heat-resistant Betula platyphylla is a is a heat resistant variety of Betula platyphylla,which grows well in North China.In this study,the disinfecting of explants in stem segments and leaves,the differentiation medium and rooting medium were screened.The optimal differentiation medium and differentiation medium were WPM+0.5 mg/L 6-ba+0.1 mg/L IBA,with the differentiation efficiency reaching 94%,and MS+1 mg/L 6-ba+0.1 mg/L IBA,with the differentiation efficiency reaching 92%,respectively.The best rooting medium was 1/2MS+0.2 mg/L IBA with the rooting rate up to 96.7%,and the best WPM+0.3 mg/L IBA with the rooting rate up to 100%.In this experiment,the efficient regeneration system of Heat-resistant Betula platyphylla was established,which is of great significance to the garden popularization and application of efficient propagation of seedlings.
