鞑靼忍冬和疏花蔷薇组培快繁体系的建立研究

摘    要:以西天山野果林中的重要伴生种鞑靼忍冬和疏花蔷薇带芽茎段为外植体,研究不同培养基及激素配比对芽分化、增殖及再生的影响。结果表明:腋芽诱导的最适宜培养基为MS培养基,鞑靼忍冬和疏花蔷薇诱导率分别可达99.12%和100%;适合鞑靼忍冬腋芽增殖的培养基为MS+6-BA 2.0 mg·L-1+NAA 0.2 mg·L-1+GA 31.0 mg·L-1,增殖系数可达5.37;而疏花蔷薇腋芽增殖的最佳培养基为MS+6-BA 1.0 mg·L-1+NAA 0.2 mg·L-1+GA 31.0 mg·L-1,增殖系数高达3.0;适合鞑靼忍冬组培苗生根的最佳培养基为1/2 MS+NAA 1.5 mg·L-1+6-BA 0.05 mg·L-1+活性炭1.0 g·L-1,生根率为100%;而疏花蔷薇组培苗生根最佳的培养基为1/2 MS+NAA 1.5 mg·L-1+6-BA 0.1 mg·L-1+活性炭1.0 g·L-1,生根率为66.67%。移栽到单独的珍珠岩基质中的鞑靼忍冬和疏花蔷薇组培苗成活率最高,成活率分别达90.00%和75.00%。

Abstract:In this study,the effects of different medium and hormones combination on bud differentiation,proliferation and regeneration of important auxiliary species,Lonicera japonica L.and Rosa laxa Ratz.,were investigated.The results showed that MS medium was the most suitable medium for axillary bud induction,and the induction rates of L.japonica and R.laxa were 99.12%and 100%respectively.The culture medium suitable for axillary bud proliferation of L.japonica was MS+6-BA 2.0 mg·L-1+NAA 0.2 mg·L-1+GA 31.0 mg·L-1,with the proliferation coefficient being 5.37,while the culture medium suitable for axillary bud proliferation of R.laxa was MS+6-BA 1.0 mg·L-1+NAA 0.2 mg·L-1+GA 31.0 mg·L-1,with the proliferation coefficient being 3.0.Additionally,The optimum mediums for the rooting of L.japonica and R.laxa were 1/2 MS+NAA 1.5 mg·L-1+6-BA 0.05 mg·L-1+activated carbon 1.0 g·L-1 and 1/2 MS+NAA 1.5 mg·L-1+6-BA 0.1 mg·L-1+activated carbon 1.0 g·L-1 respectively,and the rooting rates of the two kinds of plants were 100%and 66.67%respectively.Also,The survival rates of the two auxiliary species planting in perlite were 90.00%and 75.00%respectively.
