林下参叶片愈伤组织的诱导

摘    要:为建立林下参组培快繁技术体系,本研究以林下参叶片为外植体探讨消毒方式、外源激素浓度和不同年生对愈伤组织诱导的影响。结果表明:采用75%酒精1min,2%NaClO 20 min的消毒方式消毒,外植体污染率低且愈伤组织诱导率高;林下参愈伤组织最佳诱导培养基为:MS+2,4-D 1.5 mg/L+BA 0.5 mg/L+水解乳蛋白1g/L;不同年生叶片的愈伤组织诱导率没有显著差异。本研究可为林下参资源的可持续利用奠定方法基础。

Abstract:In order to establish the rapid propagation system of Panax ginseng C.A.Meyer,the effects of disinfection method,hormone concentration and different ages on callus induction were investigated by using the leaves as explants.The results showed that the explant contamination rate was low and the callus induction rate was high with the disinfection method of 75%alcohol and 2%NaClO for 1min and 20min respectively.The optimal induction medium for callus of panax ginseng was MS+2,4-D 1.5 mg/L+BA 0.5 mg/L+hydrolyzed protein 1g/L.There was no significant difference in the rate of callus induction in leaves of different ages.This study can lay a method foundation for the sustainable utilization of panax ginseng resources.
