棉花抗棉铃虫性室内生物测定棉叶活力保鲜技术研究

摘    要:在长14 cm×宽10 cm×高6 cm规格的聚丙烯盒内填聚氨酯并划3个2 cm长等距离刀口,注入20 mL纯净水后插入保鲜测试棉叶,在室温26℃和自然光周期条件下,保鲜30 d,棉叶鲜润如初,并能生根,具有很强的生命活力,从而解决了国内抗棉铃虫性测试上棉叶保鲜的难题。对197份材料的591个试剂盒抗棉铃虫性测定结果,幼虫存活率与3龄以上比率相关系数r=0.87320148,回归系数b=0.79296385,回归系数标准误=0.03185735。采用这种保鲜方法后在抗虫性测试中不需要更换棉叶和转接虫体,保障了测定的准确性。

Abstract:Filling the polypropylene box which is 14 cm×10 cm×6 cm with polyurethane,the polyurethane is delimited by three cutting edges,the length of each edge is 2 cm,and the distance between two edges is of equidistance.After injecting 20 mL pure water into three edges,inserting the tested fresh cotton leaf into the edge,and raising them under the condition of room temperature 26 ℃ and natural photoperiod.After 30 days,the cotton leaves are still fresh as that in the beginning,and they can root and keep vitality.So this fresh-keeping method can truly solve the difficult problem of keeping freshness of cotton leaf in test of resistance to Helicoverpa armgera in China.The results of the resistance of 197 materials to Helicoverpa armgera tested by 591 boxes indicated that the correlation coefficient between larvae survival percentage and rate of over 3 instar larvae was 0.87320148,the regressive coefficient was 0.79296385,the standard deviation of regressive coefficient was 0.03185735. Because this method did not need to replace the cotton leaf and transfer polypite in the test,the accuracy of insect-resistance test for cotton leaf can be ensured.
