外源激素对银杏叶中黄酮类化合物积累的影响

摘    要:【目的】通过对银杏叶片分别喷施不同的外源激素,探索促进银杏叶片黄酮类化合物积累的方法,并确定不同外源激素适宜的施用浓度和施用时间。【方法】以10年生实生银杏为试验材料,选用赤霉素(GA)、α-萘乙酸(NAA)、6-苄氨基嘌呤(6-BA)和多效唑(PP333)4种激素,每种激素设置3个浓度梯度,以乙醇溶液为对照,采用完全随机试验设计,在营养生长旺盛期(2019年5月9日)进行叶片喷施处理。每种处理9株,每隔5 d采集样叶,共采集4次。测定样叶的槲皮素、山奈酚、异鼠李素含量。【结果】不同处理间银杏叶黄酮类化合物含量存在显著差异。6-BA、GA、NAA和PP333对提高银杏叶槲皮素含量较适宜的溶液质量浓度分别为20、50、80和100 mg/L,对提高银杏叶山奈酚含量较适宜的溶液质量浓度分别为50、50、80和50 mg/L,对提高银杏叶异鼠李素含量较适宜的溶液质量浓度分别为20、80、50和100 mg/L溶液,对提高银杏叶总黄酮含量较适宜的溶液质量浓度分别为20、50、20和100 mg/L溶液。喷施激素6-BA、GA、NAA和PP333后,总黄酮浓度最高时期分别为喷施后15、20、15和20 d时,与对照相比,叶中总黄酮含量分别增加了26.2%、29.8%、48.5%和20.9%。【结论】在本试验中环境下,6-BA、GA、NAA和PP333提高银杏叶黄酮类化合物含量的最适宜质量浓度分别是20、50、20和100 mg/L。

Abstract:【Objective】In order to find some ways to promote accumulation of flavonoids in Ginkgo biloba leaves,and to determine appropriate hormone concentrations and applying time of different exogenous hormones,different exogenous hormones were sprayed on G.biloba leaves.【Method】Ten-year-old seedling G.biloba trees were selected as test materials.Used four hormones were gibberellin(GA),α-naphthalene acetic acid(NAA),6-benzaminoprine(6-BA)and paclobutrazol(PP333).Three concentration gradients were set for each hormone,and ethanol solution was used as control check treatment.Adopting a completely randomized test plan,the solutions were sprayed on leaves during the peak vegetative growth period(May 9,2019).There were 9 biological replicates per treatment.Some leaves were collected every 5 d,total four times.Contents of quercetin,campherol and isorhamnetin in sample leaves were determined.【Result】Contents of flavonoids are significantly different between different treatments.For increasing quercetin content in G.biloba leaves,suitable solution mass concentrations of 6-BA,GA,NAA and PP333 are 20,50,80 and 100 mg/L,respectively.For increasing campherol content in G.biloba leaves,suitable solution mass concentrations of four hormones are 50,50,80 and 50 mg/L,respectively.For increasing isorhamnetin content in G.biloba leaves,suitable solution mass concentrations of four hormones are 20,80,50 and 100 mg/L,respectively.For increasing total flavonoids content,suitable solution mass concentrations of four hormones in G.biloba leaves are 20,50,20 and 100 mg/L,respectively.Through spraying 6-BA,GA,NAA and PP333,peak stages of flavonoids content appear after 15,20,15 and 20 d,respectively.Compared with the control check,total flavonoids contents in leaves are increase by 26.2%,29.8%,48.5%and 20.9%,respectively.【Conclusion】In the test environment,the optimum mass concentrations of 6-BA,GA,NAA and PP333 are 20,50,20 and 100 mg/L,for increasing flavonoids content in G.biloba leaves.
