摘    要:采用改进的镰刀菌选择性培养基和产孢培养基对采自大田、苗圃耕层土壤中的镰刀菌进行单孢分离 ,共分离出 5 0个镰刀菌菌株 ,而且改进的镰刀菌选择性培养基对目标菌选择性强 ,产孢量大。用改进的SDS -SEAVG分离法和快速抽提法提取镰刀菌基因组DNA ,结果显示 ,用改进的镰刀菌DNA抽提方法抽提的DNA分子量大于 5 0kb ;用快速抽提方法获得的镰刀菌单孢系DNA可用于PCR扩增。

Abstract:Fifty isolates of% Fusarium% were recovered from topsoil of crop field and nursing bed with the selective media which were improved. The results showed that the selective media for isolating %Fusarium% were not contaminated, and the media for producing conidiospore have produced a bigger yield. Genomic DNA were extracted from the single conidial strains of %Fusarium% using improved SDS-SEAVG method and rapid method respectively. Agarose elecphresis demonstrated that the molecular weight of DNA which were extracted from the single conidial strains %Fusarium% using improved method was more than 50kb. Most of the obtained DNA using the rapid method were suitable for PCR amplification.
