根癌农杆菌介导的苦瓜遗传转化

摘    要:用含质粒载体pB I121的根癌农杆菌转化“南屿”苦瓜子叶节,通过对gus基因瞬时表达情况进行观测,对根癌农杆菌介导的苦瓜遗传转化体系相关因素进行研究.结果表明,苦瓜子叶节必须预培养3 d左右,用D600nm值为0.3-0.5之间的菌液浸泡,侵染时间为10 min,然后共培养3-4 d.再生植株经PCR检测初步证明gus基因已成功整合到苦瓜基因组中.

Abstract:The cotyledon nodes of Nanyu balsam pear were used as the receptors for transformation by Agrobacterium tumefaciens containing pBI121.By detecting the transient expression of gus gene,this experiment investigated the correlative factors affecting Agrobacterium-mediated transformation of balsam pear.The optimum conditions for the transformation were as follows: preculture duration about 3 days,bacterium density of D600 nm 0.3-0.5,infection duration 10 min,and coculture duration 3-4 days.The presence of gus gene was detected by PCR.
