摘    要:以野生鹿药(Smilacina japonica A.Gray)的根茎及根为对象,研究了鹿药多糖的超声提取工艺并测定了多糖含量。结果表明:水为提取剂,超声提取的最佳工艺参数为料液比1∶20(g∶mL),温度30℃,超声时间20 min。多糖含量测定采用苯酚-硫酸法,葡萄糖为对照品,最大吸收波长490 nm,回归方程为A=13.049C+0.003 7,r=0.999 8,该方法的精密度、重复性、加样回收率、稳定性的RSD分别为3.80%、4.43%、0.04%、0.16%。采用最佳工艺提取和含量测定,测得鹿药中多糖含量为(23.79±0.02)%。

Abstract:The rhizome of S.japonica is a folk traditional medicine.Flavonoids and saponins were isolated from S.japonica,and results of previous studies showed that the content of polysaccharide was higher.Extraction technology and content determination of polysaccharides in S.japonica were studied.The results showed that the optimum technological parameters for ultrasonic extraction were water as extraction agent,solid-liquid ratio 1 ∶ 20(g ∶ mL),temperature 30 ℃,extracting for 20 min.The polysaccharide content was determined by phenol-sulfuric acid colorimetric method,using glucose as reference substance,and the maximum absorption wavelength was 490 nm.The precision,reproducibility,recovery and stability of the proposed method were 3.80%,4.43%,0.04%,0.16%,respectively.By the extraction method and content determination method,the content of polysaccharide in S.japonica was (23.79±0.02)%.
