牡丹花瓣脂氧合酶测定方法的研究

摘    要:采用紫外分光光度法,以亚油酸钠为反应底物,研究了牡丹花瓣脂氧合酶(LOX)的酶学特性并建立了其测定体系。结果表明,牡丹花瓣脂氧合酶反应的最适pH值为4.5,最适温度为45℃,最适底物浓度为0.594 mmol/L。该酶有着较好的热稳定性,90℃保温1 h后仍保持较高活性。

Abstract:The enzymatic characteristics of lipoxygenase(LOX) in peony petal were studied by UV-spectrophotometry with the sodium linoleate as the reaction substrate,then the determination system was built.The results showed that the optimum pH value for the reaction was 4.5;the optimum temperature was 45℃;the optimum substrate concentration was 0.594 mmol/L.The enzyme had a good thermal stability.The enzyme activity kept higher in 90℃ for one hour.
