摘    要:基因修饰猪作为一种理想的动物模型已应用于生物医学研究。构建基因修饰猪的方法很多,包括原核注射、慢病毒转染、精子介导和体细胞核移植等,其中体细胞核移植具有显著优势,一直是构建基因修饰猪的重要方法。近几年,随着转座子、设计核酸酶等基因修饰新技术以及诱导性多能干细胞研究的不断进展,将进一步凸显体细胞核移植技术在基因修饰猪构建中的价值。本文比较了猪基因修饰的主要方法,并论述体细胞核移植技术在猪基因修饰中的优势、发展历程和最新应用等。

Abstract:Genetic modified pig as an ideal animal model has been widely applied to biomedical research. There are many methods to produce genetic modified pig, including pronuclear injection, lentivirus transfection, sperm-mediated gene transfer and somatic cell nuclear transfer (SCNT). SCNT has been considered as an important method for producing transgenic pig due to its advantages. Recently since the advent of many gene targeting tools including transposons, designer nucleases and induced pluripotent stem cells, SCNT become more meritorious for the production of genetic modified pig. The advantage, history and application of SCNT for genetic modification pig were reviewed.
