葡萄籽中多酚类物质提取工艺研究

摘    要:[目的]为今后葡萄籽的开发利用提供参考数据。[方法]选取提取试剂浓度、料液比、提取时间、提取温度和提取次数作单因素试验,在此基础上进行正交试验,优化葡萄籽中多酚类物质的提取工艺。[结果]随着甲醇体积分数的提高、料液比的增加、温度的升高及提取时间的延长,葡萄籽中多酚含量均增加。随着提取次数的增加,葡萄籽中多酚含量逐渐减少。各因素对葡萄籽多酚含量的影响程度从高到低依次为提取温度>甲醇体积分数>提取时间>料液比。[结论]葡萄籽中多酚类物质的最佳提取工艺为:体积分数70%的甲醇,料液比1∶20(g/ml),90℃水浴,浸提30min提取次数为2次。

Abstract:[Objective] The aim of the research was to provide the reference basis for the development and utilization of grape seeds in future.[Method] The concn.of extracting reagent,the material-liquid ratio,extracting time,extracting temperature and extracting times were selected to make the single factor experiments.On this basis,the orthogonal test was made to optimize the extraction technology of polyphenol substances from grape seeds.[Result] With the enhancing of volume fraction of methyl alcohol,the increasing of the material-liquid ratio,the increasing of temperature and the prolonging of the extracting time,polyphenol content in grape seeds was all increased.With the increase of extracting times,polyphenol content in grape seeds gradually was decreased.The effects of each factor on polyphenol content in grape seeds from high to low were as follows:extracting temperature > volume fraction of methyl alcohol >extracting time > the material-liquid ratio.[Conclusion] The optimum extraction technology of polyphenol substances in grape seeds was as follows:the volume fraction of methyl alcohol of 70%,the material-liquid ratio of 1∶20,water bath at 90 ℃ and extracting for 30 min.
