微波联合大孔树脂提取腊梅花中总黄酮的研究

摘    要:目的]对微波联合大孔树脂提取腊梅花总黄酮的新工艺进行研究。方法]在不同的提取工艺条件下进行平行试验、正交试验和对比试验。结果]原料0.500 g的腊梅花粉末,以70%的乙醇溶液作提取剂,提取剂的用量为1∶60(g/ml),微波功率为288 W,在提取时间为450 s下提取次数3为最佳工艺。确定AB-8为腊梅花总黄酮的吸附树脂。其最佳静态吸附工艺条件为:温度30℃,吸附时间3 h;最佳动态解吸工艺条件为:流速2.6 ml/min,上样液pH值4.3,30 ml 70%乙醇洗脱。与溶剂浸提法相比,采用微波法提取腊梅花总黄酮提取时间由20 h缩短为450 s,提取率从79.86%提高到92.23%,而且AB-8分离腊梅花总黄酮分离效果明显,成本低。结论]微波联合大孔树脂提取腊梅花总黄酮可工业化推广。

Abstract:Objective] The research aimed to study a new method of microwave joint macroreticular resin extracting flavonoids from Chimonanthus praecox(L.) Link..Method] Under different extraction conditions,the authors made parallel test,orthogonal test and contrast test.Result]The optimal condition was as follow: Raw materials was 0.500 g Chimonanthus praecox(L.) Link.dry powder,the extraction agent was 70% alcohol solution,the ratio of raw material to extraction agent was 1∶60(g/ml),the microwave power was 288 W,and the extraction time was 450 s for three times.AB-8 was determined as the absorption resin to total flavonoids of Chimonanthus praecox(L.) Link..The best passive absorption process conditions was as follow: Temperature was 30 ℃,adsorption time was 3 h.The best dynamic desorption process was as follow: Velocity was 2.6 ml/min,pH value of the upper liquid was 4.3,30 ml of 70 percent ethanol was used to clear.Compared with solvent extraction,the extraction time of microwave extraction was reduced from 20 h to 450 s,the extraction rate was increased from 79.86% to 92.23%,and that AB-8 was used to extract Chimonanthus praecox(L.) Link.flavonoids were very effective and low cost.Conclusion] microwave joint macroreticular resin extraction flavonoids from Chimonanthus praecox(L.) Link.can promote industrialization.
