烷化剂EMS处理水稻愈伤组织诱导突变的方法初探

摘    要:采用EMS 处理水稻体细胞愈伤组织诱导变异,试验表明,采用浸渍法处理2h ,EMS 对水稻体细胞愈伤组织的半致死质量浓度为1 ~5 g/L;采用滴加法处理,每管加0 .1 ml 时,半致死浓度为50 g/L左右。根据出苗率和变异情况,用滴加法处理,每管滴加0 .2 ml 时,处理浓度以1 g/L为好。经EMS 处理后,再生植株株系的变异频率比对照高5 .02 % 。

Abstract:The method and optimum concentration that EMS was used to treat rice somatic callus for mutant induction and some mutant lines were obtained. The result showed that the concentration of 1~5m g/L 50%LD of EMS was used to soak rice somatic calli for two hours; and in dropping method, the concentration was 1~5m g/L but 0.1ml EMS solution was added on the surface of callus every tube. According to the result of mutation of generation the concentration of 1m g/L was appropriate. The mutation ratio of generation was significantly higher than check after EMS treatment.
