籼稻成熟胚愈伤组织培养影响因素研究

摘    要:以 7种籼稻恢复系、保持系为材料 ,研究了影响成熟胚愈伤组织诱导的相关因素。结果表明 ,适当的低、高温预处理可以明显提高出愈率 ;CC培养基是适宜于多数籼稻愈伤组织诱导的培养基 ;不同的材料要求的 2 ,4 -D水平不一致 ,但 2mg/L的 2 ,4 -D能满足多数材料的需要 ;添加一定的脱落酸 (ABA)可以改善愈伤组织质量 ;继代时间因材料而异 ,但多数以 1 2d为宜 ;适当浓度的脯氨酸 (Pro)、二甲基亚砜 (DMSO)、谷氨酰胺 (Gln)能显著的降低褐化率 ,硝酸银的作用不明显 ;通过培养基轮换、2 ,4 -D和ABA的浓度调节以及对愈伤组织干燥培养、高渗培养等逆境调节 ,可以在一定程度上控制愈伤组织的状态 ,保持良好的胚性

Abstract:Some factors which were involved in mature embryos callus culture in Indica Rice were studied in this experiment by using 7 varieties. The results indicate that both cold and hot pretreatment can greatly improve the frequency of callus induction. CC culture media is tested to be the best media for mature embryos callus induction. Different materials are different in the demands of 2,4-D, but the concentration at 2?mg/L can commonly get satisfied results on induction. At the same time, another hormone ABA can be used to improve the condition of the callus. It is also found that the best replacing period of callus for almost all materials is about 12 days. Some additives, such as Pro, DMSO, Gln were tested can obviously decrease the browning rate of callus, but the effect of AgNO_3 is not investigated. At last, we can also maintain and improve the embryogenesis ability of callus by crossing culture media of NB and MS, by controlling the concentration of hormone 2,4-D and ABA, and by creating circumstances through dry culture and permeating threaten treatment.
