强光对鲻鱼视网膜的光化学损伤的初步研究

摘    要:给鲻鱼以2 000 lx光照12 h,采用酶学分析的方法,比较了鲻鱼在强光光照前和光照后1、37、d的视网膜内一氧化氮合酶(NOS)酶活力和丙二醛(MDA)含量。结果表明:对照组视网膜内NOS酶活力为(0.106 5±0.015 36)U/mgprot,3个实验组视网膜内NOS酶活力分别为(0.157 8±0.010 84)(、0.126 0±0.017 83)(、0.132 8±0.023 08)U/mgprot,光照后的实验组NOS酶活力显著高于光照前的对照组(P<0.05);对照组视网膜内MDA含量为(0.077 4±0.011 85)nmol/mgprot,3个实验组视网膜内MDA含量分别为(0.125 2±0.013 69)(、0.115 7±0.017 87)(、0.101 6±0.009 12)nmol/mgprot,光照后实验组MDA酶活力极显著高于光照前的对照组(P<0.01)。该实验结果与大鼠、兔等哺乳动物光损伤后视网膜内NOS酶活力和MDA含量变化趋势相似,推断鲻鱼在强光照射后视网膜会产生光损伤,为进一步研究光诱围网船作业对海洋中上层趋光鱼类产生影响打下了基础。

Abstract:The Mugil cephalus were exposed in 2 000 lx for 12 hours,the activity of the nitric oxide synthase(NOS) and the content of MDA were measured in retina of the Mugil cephalus when they were collected before light-induced and after light-induced.The Mugil cephalus were divided into the control group and experimental groups.The exeperimental groups were exposed in the strong light,and they were collected in 1,3,7 days after exposed in the strong light.The activity of NOS in the control group's retina was(0.106 5±0.015 36) U/mgprot,and the experimental groups were(0.157 8±0.010 84),(0.126 0±0.017 83),(0.132 8±0.023 08)U/mgprot respectively.The levels of NOS in the experimental groups were significantly higher than in the control group(P<0.05).The content of MDA in the control group's retina was(0.077 4±0.011 85) nmol/mgprot,and the experimental groups were(0.125 2±0.013 69),(0.115 7±0.017 87),(0.101 6±0.009 12)nmol/mgprot respectively.The levels of MDA in the experimental groups were greatly significantly higher than in the control group(P<0.01).The results were similar with those of the rats and rabbits'retina when they were exposed in the strong light.It was concluded that the retina of Mugil cephalus was light-injured probably,which can be of great importance to refer to the impacts on light-purse seine fisheries.
