鲤脑垂体和HCG诱导雌性日本鳗鲡性腺成熟过程中血清生化成分的变化

摘    要:使用鲤脑垂体和HCG对雌性日本鳗鲡人工催熟,用临床医学检验方法,测定分析了雌性日本鳗鲡卵巢发育过程中,血清总蛋白(TP)、血清甘油三脂(TG)、血清胆固醇(TC)、血糖(Glu)、血钙(Ca)和血清无机磷(P)6项生化指标的水平及变化趋势,并与对应的不同卵巢发育阶段的性腺成熟系数(GSI)进行比较分析。结果表明,84d的注射过程中,实验组的性腺逐渐成熟,GSI显著升高,第12针时达到46.32%±1.75%;而对照组性腺GSI一直在2.42%±0.18%。实验组和对照组血清TP水平相对平稳,各组变化无显著差异(P>0.05)。实验组和对照组血脂在56d前差异显著(P<0.05):实验组血清TG,TC一直维持较高水平,平均分别为(12.67±2.83)mmol/L和(15.54±3.01)mmol/L;对照组TG和TC一直下降且水平较低,平均分别为(6.33±4.14)mmol/L和(8.53±3.30)mmol/L。实验组和对照组血清Glu均在实验初的较低水平基础上持续上升。相关分析表明,实验组血清TG与GSI水平极显著负相关(r=0.732,n=6,P<0.01)。实验组血清Ca与GSI(r=0.961,n=6,P<0.01),血清P与GSI(r=0.775,n=6,P<0.01)都呈现出极显著的相关性:血清Ca和P在28d后持续上升,GSI逐渐增大。对照组中的血清Ca和P水平则一直维持在一个较低的水平。研究结果表明:雌性日本鳗鲡卵巢发育与其脂类代谢密切相关,无机离子(Ca,P)在此过程中起到重要作用。利用繁殖鱼体营养代谢特点,检测日本鳗鲡血脂或血清Ca和P水平,可成为初步判断雌性日本鳗鲡发育情况的新方法。

Abstract:Based on the gonadosomatic indexes (GSI) of different developmental ovary stages of the fe-male Japanese eel during carp pituitary extract and HCG induced maturation, the paper measured and analyzed six biochemical components of the serum, namely total protein (TP), triglycerides (TG), total cholesterol (TC), blood glucose (Glu), calcium (Ca) and phosphorus(P) by medical inspection methods. The results showed that the gonad of treatment group got matured gradually during 84 days with GSI in-creasing significantly to 46.32%±1.75% at the 12th injection; while GSI of the control group stayed at 2.45% ± 0.30%. There was no difference between the TP level of treatment group and control group (P > 0.05) which remained relatively stable. However, the blood lipid displayed significant difference (P < 0.05) until 56th day: TG and TC of the treatment group maintained a high level, averaged at (12.67 ± 2.83) mmol/L and (15.54 ± 3.01) mmol/L respectively; while control group kept a low level, averaged at (6.33 ± 4.14) mmol/L and (8.53 ± 3.30) mmol/L respectively. The Glu level of both groups carried on rising from a low level at the beginning. Correlation analysis indicated that there was significant negative correlations between TG and GSI in treatment group (r =?0.732, n=6, P < 0.01). Significant correlations were found between Ca and GSI (r =0.961, n=6, P < 0.01), P and GSI (r =0.775, n=6, P < 0.01)in treatment group: as the growth rates of GSI, and the level of Ca and P increased dramatically from 28th day, while the control group stayed at a low level. It turned out that there is a close relationship between ovary development and lipid metabolism, and some inorganic ions (Ca, P) have played an important role in this process. Accord-ing to the nutrition requirement and metabolic rule of the female Japanese eel, it’s a new potential way to judge ovary developmental stages by measuring and analyzing the level of blood lipid or serum Ca and P.
