泽泻根茎无性系建立的研究

摘    要:为保存野生根茎大的泽泻种质,以根茎为材料,成功地建立起泽泻无性系。结果证明:MS+BA0.3 mg/L+NH4H2PO4 50 mg/L+2,4-D 1.6~2.0 mg/L是愈伤组织诱导和继代培养的适宜培养基;MS+NAA 0.1 mg/L+AgNO3 0.5 mg/L+GA3 1.0 mg/L+BA 0.9 mg/L是愈伤组织分化培养的适宜培养基;1/2MS+IAA 0.2 mg/L+NAA 0.1 mg/L的液体培养基是泽泻不定芽生根培养的适宜培养基;在温室中试管苗易移栽成活,移植到材料原产地的试管苗生长旺盛,保持了根茎大的性状。

Abstract:To protect the wild germplasm of Alisma orientale with the big rhizome,its asexual line was established with the rhizome as material.The results showed that the ideal medium for callus induction and subculture was MS + BA 0.3 mg/L + NH4H2PO4 50 mg/L + 2,4-D 1.6 ~ 2.0 mg/L;the ideal medium for callus differentiation was MS + NAA 0.1 mg/L + AgNO3 0.5 mg/L + GA3 1.0 mg/L + BA 0.9 mg/L;the optimum medium for rooting of adventitious buds was the liquid medium with 1 /2 MS + IAA 0.2 mg/L + NAA 0.1 mg/L.The tube seedlings could be transplanted easily in the greenhouse,and grew vigorously with the big rhizome when transplanted to the origin.
