甘肃陇南地区小麦条锈菌群体遗传多样性分析

摘    要:为了解甘肃陇南地区小麦条锈菌群体的遗传多样性,采用TP-M13-SSR荧光标记技术,对该地区8个种群202个小麦条锈菌分离株基因组DNA进行SSR标记分析。结果表明,甘肃陇南地区小麦条锈菌群体遗传多样性比较丰富。在物种水平上,观察等位基因数(Na)为1.88,有效等位基因数(Ne)为1.50,Nei’s基因多样性指数(H)为0.30,Shannon信息指数(I)为0.46,多态位点百分率(P)为87.5%;种群之间遗传多样性有显著差异。中梁种群、秦安种群的遗传多样性相对较高,武都种群、文县种群、齐寿种群和平南种群次之,礼县种群、西和种群遗传多样性相对较低。AMOVA分析结果表明,小麦条锈菌群体间和群体内都存在着一定的遗传分化,群体间的遗传变异占总变异的4.05%,群体内遗传变异占95.05%。

Abstract:The population genetic diversity of P.striiformis f.sp.tritici was investigated among 202 isolates collected from 8 different regions of Longnan,Gansu province with TP-M13-SSR analysis.The results showed that the average number of alleles(Na) per locus was 1.88,and the effective number of alleles(Ne) was 1.50 among the Longnan population.The Nei’s gene diversity(H) and Shannon’s information index(I) were 0.30 and 0.46,respectively.The genetic diversity among the population from Zhongliang and Qing’an was much higher than that from Wudou,Wenxian,Qishou and Pingnan.The genetic diversity among the population from Lixian and Xihe was the least.Analysis of AMOVA suggested that about 4.05% of the total variation were contributed by the variations among the populations from different regions,and 95.95% of the total variation were presented within the populations.
