摘    要:对Folin-Denis法和Folin-Ciocalteus法测定水果及其制品中总多酚含量进行了研究。Folin-Ciocalteus法比Folin-Denis法更加稳定、灵敏和准确。经过一系列优化实验,提出Folin-Ciocalteus法的最适测定程序为1.0mL样品提取液中依次加入5.0mL水、1mL显色剂和3mL7.5%碳酸钠溶液,摇匀,显色2h,在765nm波长下测定吸光度。采用此程序,苹果、梨、香蕉、柿子、葡萄和葡萄酒等6种试材的没食子酸平均添加回收率为77.2%~112.1%,方法误差为0.8%~5.7%。

Abstract:Determination methods (Folin-Denis method and Folin-Ciocalteus method) of total polyphenols in fruit and its derived products were studied. Folin-Ciocalteus method was more suitable to be used than Folin-Denis method in stability, sensitivity, and accuracy. After a series of optimization experiments, the best determination procedure of Folin-Ciocalteus method was put forward, that is, 5.0 mL water, 1.0 mL coloration reagent, and 3.0 mL 7.5% sodium carbonate solution are orderly added into 1.0 mL extract of samples, then blend it, and measure its optical density at 765 nm after coloration for 2 hours. Under this procedure, average recovery rate of gallic acid in 6 materials (including apple, pear, banana, persimmon, grape and wine) is 77.2% to 112.1%, relative deviation of the method is 0.8% to 5.7%.
