摘    要:根据黄酮类化合物能与Al3+形成稳定的荧光络合物,建立一种测定玉米须总黄酮的新方法。用60%乙醇溶液,在功率400W下超声处理20min,提取玉米须有效成分;以芦丁为标样,选择激发波长为λex=445nm,发射波长为λem=542nm,采用荧光分光光度法测定玉米须中总黄酮含量,并进行加标回收实验,以验证方法的准确性。方法检出限为1.08×10-9mol/L,线性范围在0~0.04mg/mL之间;线性回归方程:y=216.63x+137.93,相关系数r=0.999 7,平均回收率为98.3%~100.8%,相对标准偏差(RSD)为0.88%。这种方法操作简便、快速、准确,具有良好的分析应用前景。

Abstract:A new method for total flavanoid determination in cornsilk was established based on the reaction of total flavanoid with alumimumion to form fluorescent material.The sample was extracted with 60% ethanol by ultrasonic extraction under 400W for 20min.Total flavanoid was determined by spectrofluorophotometer with rutin as standard reference at the excitation wavelength of 436 nm and the emission wavelength of 483 nm.The detection limit was 1.08×10-9 mol/L.The range of linearity with concentration of rutin was between 0~0.02 mg/mL,linearity relationship y=216.63x+137.93,correlativity 0.999 7,average rate of recovery 98.3%~100.8%,and RSD 0.88%.The method is simple,rapid and accurate with a bright future in analytical applications.
