摘    要:目的]寻找利用发酵法生产透明质酸的菌株。方法]从奶牛鼻黏膜分离到1株透明质酸产生菌,对其进行筛选和菌落生化反应鉴定。分别用紫外线诱变和亚硝基胍诱变处理该菌株,研究诱变前后透明质酸含量的变化。结果]经初步鉴定该菌株为兽疫链球菌的C群链球菌。筛选菌株的产物呈典型的HA红外吸收图谱。经诱变处理后,菌种菌落形态更大且更明显,呈黏液状。该菌株透明质酸的产量由初始菌株的3.69g/L提高到6.94g/L。结论]该研究为利用发酵法生产透明质酸提供了科学依据。

Abstract:Objective] The aim of the research was to seek the strains for producing hyaluronic acid (HA) by fermentation method. Method] A strain of bacteria for producing HA was isolated from nasal mucosa of milk cow and it was screened and identified by colony biochemical reaction. This strain was treated by ultraviolet mutation and nitrosoguanidine mutation resp. and the changes of HA content before and after mutations were studied. Result] Through the preliminary identification, this strain was C group of Streptococcus zooepidemicus. The product of the screened strain showed the typical infrared absorption spectra of HA. After mutation treatment, the colony morphology of the strain was larger and more obvious, being mucous. The HA yield of this strain was increased from 3.69 g/ L of the original strain to 6. 94 g/ L. Conclusion] This research provided the scientific basis for producing HA by fermentation method.
