黄瓜花叶病毒的RT-PCR检测

摘    要:根据黄瓜花叶病毒(CMV)的复制酶基因序列设计、合成引物,以感病组织和健康组织总RNA为模板,CMV质粒作为RT-PCR反应的阳性对照,进行cDNA合成和PCR扩增.对2002-2003年采集的98份黄瓜病毒病样本进行了检测,结果从感病组织中扩增出与预期的767bp大小一致的目标片段,而健康组织无此扩增产物.结果表明98份材料中96.94%检测到CMV.

Detection of Cucumber Mosaic Virus on Cucumber by RT-PCR
Abstract:A pair of primers were designed and synthesized based on the sequences of CMV RNA.RT-PCR was performed us-ing total RNA as template which was extracted from virus-infected and healthy cucumber leaves.A fragment of about767bp was am-plified from the infected samples,but not from healthy samples.A total of98cucumber samples associated with viruses collected from several regions of Tianjin in2002and2003were detected.The results showed that the incidence rate of cucumber mosaic viruses(CMV)was96.94%.
