| 摘 要： | 本文比较观察了用甘油和二甲基亚矾(DMSO)处理的鸡精子冷冻前后形态结构的变化,以阐明甘油的冷冻保护效果。结果表明,a.用甘油或DMSO作为冷冻保护剂,冷冻前后的畸形精子主要表现为颈部弯曲,甘油处理的精子这种颈部弯曲的畸形率明显增加(P<0.05),b.甘油处理的精子,解冻后表现为明显的头部原生质膜破裂,常伴有中段膨大和线粒体鞘裸露的现象。由此推测,甘油对鸡精子结构的不良作用可能与冷冻解冻过程中稀释液的渗透压变化有关。

| Abstract: | The morphological changes of chicken spermatozoa were observed by means of optical, scanning electron and transmission electron microscope before and after freezing with either glycerol or dimethylsulphoxide(DMSO)as protectant. The results obtained are as follows: 1. Treated with either glycerol or DMSO the abnormal spermatozoa were mainly found with a bent neck both before and after freezing,and the number of spermatozoa with a bent neck increased remarkably (P<0.05) when treated with glycerol; 2. The spermatozoa protected by glycerol usually displayed the following morphological changes after thawing: the head plasma membrane ruptured, the midpiece swollen and the mitochondria exposed. These results suggest that the morphological changes with glycerol-treated spermatozoa might be attributed to the changes in the osmotic pressure of the semen diluent in freezing and thawing processes. |
