嗜酸乳杆菌促生长物质及增菌培养基优化研究

摘    要:研究了在MRS培养基中添加不同营养物质对嗜酸乳杆菌生长的影响,进一步采用正交试验优化筛选了嗜酸乳杆菌的增殖培养基。结果表明,在MRS培养基中添加菊粉、乳清粉、酪蛋白水解物、胡萝卜汁可显著促进试验菌株的细胞生长,而乳糖、西红柿汁、啤酒的添加相对而言对菌的增殖影响不大;利用L9(34)正交试验筛选出增殖因子的最佳配比为:菊粉0.5%,乳清粉0.5%,酪蛋白水解物0.5%,胡萝卜汁5%。最后使用优化出的增菌培养基测定生长曲线,同时测定pH和滴定酸度的变化,确定增菌培养终止时间为10 h,此时活菌数可达2.59×1010cfu/mL,较对照MRS培养基的活菌数提高了22.72倍。

Abstract:Effects of different nutriments on the growth of Lactobacillus acidophilus were examined.The results showed that inulin,whey powder,casein peptone,carrot juice medium can promote the growth of Lactobacillus acidophilus to some extent,of which whey powder has the best promotio effect.The optimum growth promoting factors were as follows:0.5% of inulin,0.5% of whey powder,0.5% of casein peptone,5% of carrot juice.Growth curve with optimized stimulated culture were determined.According to the growth curve,the terminal time was at 10 hour,and the viable counts of bacteria got to 2.59×1010 cfu/mL.
