Abstract: | AIM: To investigate the effects of miRNA-specific inhibitors on proliferation of gastric cancer cells.METHODS: The specific 2’-methoxy-modified RNA oligonucleotides (miRNA inhibitors) of miR-17, miR-21, miR-106a, and miR-421 were designed, synthesized and transfected into SGC-7901 and MGC-803 gastric cancer cells by liposome. The expression of miRNAs in cells was detected by quantitative real-time reverse transcriptase-polymerase chain reaction. Finally, the cell proliferation was analyzed by MTT assay.RESULTS: Four miRNA-specific inhibitors significantly inhibited the expression of corresponding miRNAs in SGC-7901 and MGC-803 cells. miRNA inhibitors of miR-17, miR-106a, and miR-421 showed the inhibitory effect of proliferation on gastric cancer cells in a dose-dependent manner. The growth inhibitory effect on SGC-7901 cells was better than that on MGC-803 cells.CONCLUSION: The miRNA-specific inhibitors effectively inhibit the proliferation of gastric cancer cells and provide a new method for investigating the pathogenesis of gastric cancer. |
