独蒜兰离体快繁技术研究

摘    要:以野生独蒜兰成熟未开裂的蒴果为外植体,对独蒜兰离体快繁技术进行研究。结果显示:用清水冲洗后再用75%酒精消毒30s,0.1%升汞消毒10min,污染率低至26%。培养基VW+0.005mg·L^-1TDZ+0.3mg·L^-1NAA适宜于独蒜兰的种子萌发培养,在该培养基中独蒜兰种子的萌发率为39.3%;适宜独蒜兰芽增殖的培养基为VW+1.0mg·L^-16-BA+0.2mg·L^-1NAA,在该培养基中独蒜兰芽的增殖系数为7.6;在VW+0.2mg·L^-1NAA+0.2mg·L^-1IBA+0.3g·L^-1活性炭的培养基上,独蒜兰的生根率为92%。独蒜兰组织培养技术体系的建立,对于解决其生产用苗紧缺以及遗传改良有着重要的理论价值及实践意义。

Abstract:Using the wild Pleione bulbocodioides (Franch.)Rolfe mature and unbroken capsule as explant,to study the tissue culture techniques of it.The result showed that:rinsed with water,then disinfected with 75% alcohol for 30 s and 0.1% mercuric chloride for 10 min,the contamination rate was 26%.VW+0.005 mg · L^-1 TDZ+0.3 mg · L^-1 NAA was suitable for P.bulbocodioides seed germination culture,the germination rate was 39.3%;VW+1.0 mg · L^-1 6-BA+0.2 mg · L^-1 NAA was suitable for P.bulbocodioides proliferation culture,the proliferation multiple was 7.6;In culture medium of VW+0.2 mg · L^-1 NAA+0.2 mg · L^-1 IBA+0.3 g · L^-1 activated carbon,the rooting rate was 92%.The technique establishing of P.bulbocodioides tissue culture have the important thoeretical and practical significance on solving the problem of insufficient supply of P.bulbocodioides in the production and genetic modification.
