大豆花叶病毒抗血清制备和应用方法研究

摘    要:用差速离心法提纯6个大豆花叶病毒株,提纯液用紫外分光光度计扫描,以260mm的OD值计算病毒汁液浓度.影响提纯病毒浓度的因素有:(1)不同毒株同一提纯方法得率差异很大;(2)采病叶的时间:症状最明显时采叶提纯得率最高;(3)PEG处理时间以3.5小时较好. 将提纯病毒免疫兔子(以6号兔为例),共注射8.34mg,心脏采血,取得抗血清,通过微量凝聚、微量沉淀反应及琼脂扩散试验,在方法上进行了研究.


PURIFICATION AND SEROLOGICAL STUDY OF SOYBEAN MOSAIC VIRUS (SMV)
Abstract:This paper reports results of the study on preparation of antiserum to Soybean Mosaic Virus and serological test with respective antiserum. SMV was purified from infected leaves, 15-30 days after showing symptoms by chloroform-butanol clearification, followed by polyethylene glycol (PEG) precipitation and ultracentrifugation. The ultraviolet absorption spectrum of the purified SMV was measured in a UNICAM SP 800 Spectrophotometer. Our data showed that the strains of SMV, time between leaves infected to purify, hours of PEG precipitation affected the final yield production of virus particles. In our test strain SMVy-51 failed to get high titre after 6 injections because the low concentration of virus antigen. However has reached high level of antibody soon followed the strengthening immunization. Study on storage days of diluted antisera and infected leaf soak(as antigen)affecting the titre of antisera was carried out also. Serological tests have been made with micro-agglutination and micro-precipitation gel double diffusion.
