浸提法提取橘皮中叶黄素的研究

摘    要:目的]探讨浸提法提取橘皮中叶黄素的工艺。方法]以提取配料比(25∶1、75∶2、50∶1),提取温度(30、45、60℃),提取时间(48、72、96 h)为3因素,采用正交试验设计,确定四氢呋喃的最佳提取条件,用分光光度计测定提取液在最大吸收波长下的吸光度A,确定最佳的提取溶剂和最佳提取条件。结果]从干燥柑橘皮中提取叶黄素粗品的最佳溶剂为四氢呋喃;其最佳提取条件为:提取温度60℃,提取时间72 h,物料比(柑橘皮∶四氢呋喃m/V)27∶5;确定了从浸提液浓缩精制为粗产品的工艺,采用四氢呋喃提取液过柱洗脱获得的叶黄素含量达到50%以上,表明四氢呋喃对柑橘皮中叶黄素的提取能力较强。结论]采用浸提法工艺简单,操作方便,适合农产品的粗加工。

Abstract:Objective] The study aimed to discuss the process on the extraction of lutein in orange peel.Method] With the extracting mixture ratio(25∶1,75∶2,50∶1),temperature(30,45,60 ℃),time(48,72,96 h) as 3 factors,the orthogonal experiment design was used to make sure the optimum extraction condition of tetrahydrofuran,and the absorbance A of extraction liquid was measured at the maximum absorption wavelength by spectrophotometer so as to determine the optimum extraction solvent and extraction condition.Result] The optimum solvent of extraction lutein crude product from dry orange peel was tetrahydrofuran.Its optimum extraction condition was as follows: extraction temperature of 60 ℃,extraction time of 72 h,material ratio(orange peel: tetrahydrofuran m/V) of 2∶75.The process of concentrating and refining the extraction liquid into the crude product was determined and the lutein obtained through the chromatographic column and elution by tetrahydrofuran extraction liquid could reach above 50%,which showed that the tetrahydrofuran had stronger extraction ability on lutein in orange peel.Conclusion] This extraction method was simple in process and convenient in operation,which suited for rough machining of farm products.
