摘    要:目的]建立土鳖虫中尿囊素含量的测定方法,考察不同产地土鳖虫中尿囊素的含量,为土鳖虫药材质量标准的制订提供依据。方法]采用HPLC法对土鳖虫的尿囊素进行定量分析,色谱柱为Agilent ODS-C18(4.0 mm×250.0 mm,5μm),流动相为乙腈-0.1%醋酸,梯度洗脱,流速为1.0 ml/min,柱温为30℃,检测波长为360 nm。结果]各产地土鳖虫中尿囊素含量差异较大,其中以"致信药材"土的鳖虫尿囊素含量最高。尿囊素在0.2~2.0μg范围内线性关系良好(r=0.999 9),平均回收率为97.42%(RSD=4.5%,n=5)。结论]所建立的方法准确可靠,重复性好,可用于土鳖虫药材的质量控制。

Abstract:Objective]The aim was to establish the determination methods of allantion content in Eupolyphaga seu Steleophaga in different producing areas to provide the experiment basis for formulating the quality standard of Eupolyphaga seu Steleophaga.Method]The quantitative analysis was carried out for allantion in Eupolyphaga seu Steleophaga based on HPLC method,and chromatographic column was Agilent ODS-C18(4.0 mm×250.0 mm,5 μm),the mobile phase was the mixture of acetonitrile and 0.1% acetic acid with a gradient mode at the flow rate of 1.0 ml/min;the column temperature was 30 ℃ and the detecting wavelength was 360 nm.Result]In addition,there were greater differences of allantion content in Eupolyphaga seu Steleophagain different producing areas,of which allantion content in Eupolyphaga seu Steleophaga in"Zhixin medicinal materials" was greatest.Aallantion showed a good linera relationship at the range of 0.2-2.0 μg(r=0.999 9),and the average recovery was 97.42%(RSD=4.5%,n=5).Conclusion]The established method was accurate and reproducible,and can be used for the quality control of Eupolyphaga seu Steleophaga.
