东北百合鳞片离体培养研究

摘    要:以东北百合(Lilium distichum Nakai)的鳞片为外植体进行离体培养,探讨不同外源激素水平对其小鳞苇诱导、增殖与不定根形成的影响,建立了东北百合鳞片再生体系.结果表明:MS+BA1.0mg·L~(-1)+NAA0.5~1.0mg·L~(-1)培养基的不定芽诱导和增殖效果最好,诱导率为80.0%以上,增殖系数为4.1~4.6;1/2MS+NAA0.5~1.0mg·L~(-1)或1/2MS+IBA0.1mg·L~(-1)培养基的生根效果最好,生根率为96.0%;试管苗在草炭+珍珠岩(1:1)基质中扦插成活率为96.0%.

Abstract:Bulb scales of Lilium distichum Nakai were used as explants to study the effects of hormone on bulblets inducement, multiplication and adventitious roots formation, and an excellent regeneration system was established. The results showed that the induction, differentiation and multiplication of shoots were the best in the medium of MS+BA 1.0 mg·L~(-1)+NAA0.5-1.0 mg·L~(-1), with the induction rate of 80.0% and the multiplication coefficient of 4.1-4.6. The rooting was the best in the medium of 1/2MS+NAA0.5-1.0 mg·L~(-1) or 1/2MS+IBA0.1 mg·L~(-1) with the rooting rate of 96.0%. The survival rate of plantlets could reach 96.0% in the medium of peat and pearlite (1:1).
