耐低温兼性厌氧淀粉酶产生菌Y89的筛选及酶学特性

摘    要:从石家庄污水处理厂活性污泥中筛选得到一株兼性产淀粉酶菌株Y89,经形态学、生理生化和16S rDNA序列分析,鉴定为类芽胞杆菌属 (Paenibacillus sp.)。该菌株为耐低温菌,它的最适生长温度为16℃,最适氯化钠浓度5%,最佳pH值为6.0,pH 5.0-9.5范围内均能生长, 培养72-124h为生长稳定期,在40℃进行水浴反应,该菌株酶活较高,为386.13U/ml。

Abstract:To improve methane gas production rate in anaerobic fermentation ,a facultative strain producing amylas was isolated from activated sludge of a sewage treatment plant by using of double-flat-panel screening.The morphology,the physiological biochemical characteristics and the 16S rDNA analysis of the strain showed that this organism was Paenibacillus sp..Studies on the enzymatic properties indicated that the strain was psychrotolerant,the optimum growth temperature was 16℃ ,the NaCl optimum for growth was 5%,the fastest growth occurred within pH 6.0,the strain can also grow within a range of 5.0-9.5.The stationary phase of the strain occurred after cultured between 72 h and 124 h,water bath at 40℃,the Amylase-producing displayed maximum activity (386.13 U/mL).
