胡枝子属植物40个野生居群种子蛋白谱带多样性研究

摘    要:利用SDS聚丙烯酰胺凝胶电泳技术,对采自内蒙古、河北、山西、宁夏和甘肃5个省3种2变种的40个胡枝子属植物野生居群种子蛋白进行遗传多样性研究.结果获得32条谱带,有4条谱带为稳定表达带,其余28条谱带均有不同程度的多态性,多态率90.6%,分子量108.1~10.3 kDa,各居群间遗传相似系数变异范围为0.42~1.00,遗传变异较大,多态性较高.遗传相似系数为0.581时聚为4类:第Ⅰ类包括二色胡枝子Lespedeza bicolor的3个居群;第Ⅱ类包括23个居群,即牛枝子L.potaninii的11个居群和达乌里胡枝子L.davurica的12个居群;第Ⅲ类包括牛枝子的2个居群;第Ⅳ类包括尖叶胡枝子L.hedysaroides的4个居群和细叶胡枝子L.hedysaroides var.subsericea的8个居群.其中第Ⅲ类的牛枝子2个居群单聚一类,表明与其它牛枝子居群遗传差异大.从种的水平看,绝大部分来自于相同或相似的地理生态环境的居群聚为一类.达乌里胡枝子与牛枝子亲缘关系较近,尖叶胡枝子与细叶胡枝子亲缘关系较近,这与形态学鉴定结果相一致.由此认为,胡枝子属植物是种间、种内具有丰富的蛋白质多态性的种群,可作为研究种内变异和生态型的重要依据.

Abstract:Genetic diversity of seed storage protein was estimated on 40 wild Lespedeza populations(3 species and 2 varied species) from Inner Mongolia,Hebei,Shanxi,Ningxia and Gansu provinces,using SDS-PAGE techniques.Total 32 bands were found varing in molecular weight from10.3KDa to 108.1KDa;among which,4 bands were shared by all the populations and other 28 showed(90.6%).The genetic similarity coefficient varied from 0.42 to 1.00,giving greater genetic variation and higher polymorphism.Clustering with UPGMA using genetic similarity coefficient at 0.581 showed the existence of 4 different groups.Three populations of L.bicolor were classified as the first group.The second group consisted of 23 populations,including 11 populations of L.potaninii and 12 populations of L.davurica.Two populations of L.potaninii were included in the third group.The fourth group was made up of 4 populations of L.hedysaroides and 8 populations of L.hedysaroides.var.subsericea.At species level,a majority of populations from the same or similar areas were gathered together.L.davurica was genetically close to L.potaninii;while L.hedysaroides was close to L.hedysaroides var.subsericea,indicating that cluster analysis coincided with the results of morphologic identification. Therefore,abundant protein diversity among Lespedeza species could be an important reference to intraspecies variation and ecological type research.
