铀胁迫对大豆幼苗细胞DNA损伤的彗星试验研究

摘    要:通过室内培养方法,用八氧化三铀配制浓度为100、200、400、600、800、1000μmol·L^-1的6种铀溶液和对照组培养大豆幼苗,采用彗星试验研究了铀胁迫对大豆幼苗细胞DNA的损伤情况。结果表明,铀溶液能对大豆幼苗细胞DNA造成损伤;配制的6种浓度的铀溶液均对大豆幼苗根部细胞DNA造成损伤,其中浓度为800和600μmol·L^-1的铀溶液对大豆幼苗根部细胞DNA的损伤最严重;铀浓度为1000μmol·L^-1的铀溶液对大豆幼苗茎部细胞DNA的损伤最严重;配制的6种浓度的铀溶液对大豆幼苗叶部细胞DNA的损伤不明显。

Abstract:The uranium solutions and a comparison solution were used to cultivate soybean seedlings. The uranium solutions were compounded of U3O8, and were in the concentrations of 100 μmol·L^-1, 200 μmol·L^-1, 400 μmol·L^-1, 600 μmol·L^-1, 800 μmol·L^-1 and 1 000 μmol·L^-1, respectively. A solution for comparision was prepared. The comet assay was used to determine the damages to soybean seedlings cell DNA caused by the stress from the uranium solutions. The research results showed that the uranium solutions could promote the germination of the soybean seed, that the uranium solutions have damaged the soybean seedlings cell DNA, that the uranium solutions caused the most serious damage to the soybean seedlings′root cell DNA, that the uranium solutions in concentrations of 600 μmol·L^-1 and 800 μmol·L^-1 caused the most serious damage to the soybean seedlings′root cell DNA, that the uranium solution in the concentration of 1 000 μmol·L^-1 caused the most serious damage to the soybean seedlings′stem cell DNA and that the uranium solutions caused no obvious damage to the soybean seedlings′leaf cell DNA.
