摘    要:目的

研究烟草花多酚对α-淀粉酶及α-葡萄糖苷酶的抑制作用及机制。

方法

采用酶抑制、酶动力学、荧光光谱和分子对接等试验方法对烟草花多酚的体外降糖活性及机理进行研究。

结果

烟草花多酚对α-淀粉酶及α-葡萄糖苷酶的半抑制质量浓度(IC50)分别为0.188 5 mg/mL和0.002 5 mg/mL,抑制类型均为混合型抑制,且多酚与2种酶均发生静态猝灭,分子对接显示小分子与酶蛋白之间通过疏水和氢键等作用结合而发挥抑制作用。

结论

烟草花多酚的体外降糖活性显著,具有天然酶抑制剂开发的潜力。

Abstract:PurposeTo study the inhibitory effects and mechanism of polyphenols from tobacco flower on α-amylase and α-glucosidase. MethodsThe hypoglycemic activity and mechanism of polyphenols from to bacco flower in vitro were studied by enzyme inhibition, enzyme kinetics, fluorescence spectroscopy and molecular docking experiments. ResultsThe 50% inhibiting mass concentration (IC50) of polyphenols on α-amylase and α-glucosidase were 0.1885 mg/mL and 0.0025 mg/mL, respectively; the inhibition type was mixed type, and the static quenching of polyphenols and the enzyme protein occurred. The molecular bonding showed that the inhibition activity between small molecules and enzyme protein was formed through hydrophobic and hydrogen bonding. ConclusionPolyphenols from tobacco flower have significant hypoglycemic effect in vitro and have the potential to develop natural enzyme inhibitors.
