摘    要:为探讨甘肃境内鼢鼠的系统进化关系,采用PCR技术对采集的4种鼢鼠线粒体DNA ND4序列进行扩增,经序列测定后用Clustal X1.83对比,获得了17条长度为425~438 bp不等的同源序列。通过DAN SP4.0比较分析,共检出160个多态信息位点,定义了15种单倍型,单倍型多样度为(Hd):0.980±0.024。采用MEGA4.0中的NJ法、MP法构建的分子进化树表明:高原鼢鼠两群体聚在一起后再与斯氏鼢鼠聚在一起,处于较进化的位置,后与秦岭鼢鼠聚在一起形成一进化枝,再与甘肃鼢鼠群体聚在一起,甘肃鼢鼠最先分化出来,且单独成一进化枝。

Abstract:To evaluate systematical evolution relationship and provide reasonable basis and reference for classification of Eospala,the ND4 gene of four kinds of zokors were analyzed using PCR method.Direct sequencing 17 individuals of 425~438 bp homologous nucleotide sequences were obtained by using Clustal X1.8,and it showed 160 polymorphic nucleotide sites among homologous sequence and 15 haplotypes were found.Haplotype diversity is 0.980±0.024.In addition,the molecular phylogenetic trees based on ND4 gene was constructed by NJ and MP method using software MEGA4.0,two major evolutionary lineages,Clade A(Myospalax baileyi,M.smith and M.rufescens),and Clade B(M.cansus),were recognized within the specimens.Actually,M.baileyi and M.smith clusters together with high bootstrap values,then clusters together with M.rufescens,while M.cansus remains the basal clade.
