摘 要: | [目的]研究离体培养条件下植物生长调节剂对杜梨种胚休眠解除的影响。[方法]以低温处理20 d的杜梨种胚为材料1、/2 MS为基本培养基,以培养基中不含植物生长调节剂为对照,研究植物生长调节剂对杜梨种胚休眠解除的影响。[结果]6-BA和赤霉素均有助于杜梨种胚的休眠解除,提高种胚萌发率。当培养基中添加6-BA时,在培养的第24天和第36天种胚萌发率、苗高和根长分别较对照提高18.2%和21.39%、0.86和0.16 cm、0.94和1.65 cm;当培养基中添加赤霉素时,在培养的第24天和第36天种胚萌发率、苗高和根长分别较对照提高16.86%和21.46%0、.70和0.16 cm0、.94和1.39 cm。在培养基中同时添加吲哚乙酸和活性炭效果更好。[结论]在培养基中添加植物生长调节剂有利于杜梨种胚的休眠解除。

Effect of Plant Growth Regulator on Dormancy Release of Seed Embryo of Pyrus betulaefolia in vitro Culture |

Abstract: | [Objective] The research was to study the effect of plant growth regulator on dormancy release of seed embryo of Pyrus betulaefolia in vitro culture.[Method] With the seed embryo of P.betulaefolia treated by low temperature for 20 d as tested material,1/2 MS as basic medium,and with the treatment without plant growth regulator in medium as CK,the effect of plant growth regulator on dormancy release of seed embryo of P.betulaefolia was studied.[Result] 6-BA and gibberellin were all helpful to release the dormancy of seed embryo of P.betulaefolia and improve the germination rate of seed embryo.When 6-BA was added into the medium,the germination rate,seedling height and root length were increased by 18.2% and 21.39%,0.86 and 0.16 cm,0.94 and 1.65 cm than that of CK on the 24th d and 36th d after culture.When gibberellin was added into the medium,the germination rate,seedling height and root length were increased by 16.86% and 21.46%,0.70 and 0.16 cm,0.94 and 1.39 cm than that of CK on the 24th d and 36th d after culture,and the effect was better when indoleacetic acid and activated carbon were added into the medium.[Conclusion] It was beneficial to dormancy release of seed embryo of P.betulaefolia to add plant growth regulator into the medium |
