月季花粉离体萌发液体培养基组分的优化

摘    要:以月季品种‘Sorbet fruite'的花粉为材料,通过单因子试验比较了液体培养基中蔗糖及B、Ca、Mg、K对月季花粉离体萌发的影响,以此为基础利用L_(25)(5~6)的正交试验,比较了蔗糖、Ca(NO_3)_2·4H_2O、H_3BO_3对月季花粉萌发的影响。结果表明:蔗糖、Ca(NO_3)_2·4 H_2O和蔗糖×H_3BO_3的交互作用对‘Sorbet fruite'花粉萌发有极显著影响。适宜培养液为蔗糖20%+H_3BO_3(50~100)mg/L+Ca(NO_3)_2·4 H_2O(100~200)mg/L。在适宜的培养液中加入30%的PEG-4000,萌发率和花粉管长度均有极显著增加。

Abstract:Fresh pollen of‘Sorbet fruite'was employed as experimental material,and comparisons were made for the effects of sucrose,boron,calcium,magnesium and potassium on the in vitro ger- mination of rose pollen through simple factorial experiment.And L_(25)(5~6)orthogonal design was used to compare the effects of sucrose,boron,calcium on the germination of rose pollens.Results showed that sucrose,calcium and sucrose×boron interaction have great effects on the in vitro ger- mination of rose pollen.The optimum culture solution was sucrose 20%+H_3BO_3(50-100)mg/L+ Ca(NO_3)_2·10H_2O(100-200)mg/L.The optimal culture solution supplemented with 40 % PEG- 4000 increased both the pollen germination rate and the pollen tube length significantly.
