摘 要: | 利用RT-PCR和PCR技术扩增出近期猪瘟流行野毒株的E2基因,将其克隆到载体上测定核苷酸序列,根据Alfort株和Brescia株、C-株确定起始氨基酸三联体的正确位置后推导出其氨基酸序列,结果表明近期流行的猪瘟毒株与疫苗毒株和强毒株之间存在一定差异;利用DNAstar软件分析E2基因编码的gp55蛋白的疏水性、抗原性和二级结构,并与猪瘟疫苗毒株(C-株)进行比较,结果表明,目前流行的猪瘟病毒与疫苗毒的gp55蛋白在抗原性、二级结构上存在一定的差异,而C末端疏水性差异不大。

Abstract: | The E2 gene of prevalent virulent strain of classical swine fever virus was amplified by the RT-PCR and PCR.The amplified fragments was cloned into vector and then sequenced,based on the amino-acid sequences of Alfort and Brescia strains,amino-acid sequence of gp55 protein were deduced.The results showed that there was some differences among recently prevalent virulent strains,C-strain and virulent strains.Antigenicity,hydrophobicity and secondary structure of E2 gene code gp55 protein of classical swine fever virus were analysed using DNAstar soft ware.The prevalent virulent strain and C-strain antigenicity,hydrophobicity and secondary structure were compared.The results showed that antigenicity and secondary structure were different between the prevalent virulent strain and C-strain,the C-terminal hydrophobicity has little difference between the prevalent virulent strain and C-strain. |
