摘    要: 利用RT-PCR技术从矮牵牛的紫色花瓣中克隆了两个控制花色的类黄酮3',5'羟基化酶基因Hf 1和Hf 2, 并构建了植物表达载体, 利用农杆菌介导法对铁炮百合进行了遗传转化, 获得了转基因植株。

Abstract:Flavonoid-3',5'-hydroxylase(F3',5'H) is a key enzyme in the biosynthesis of anthocyanins,which can divert the production of delphinidin pigments.Two cDNAs designated as Hf1 and Hf2 encoding F3',5'H were isolated from the purple flower of Petunia hybrida by using RT-PCR and also sequenced.A construct of Hf 2 in a sense-orientation driven by CaMV 35S promoter was constructed and transformed into Lily(Lilium longiforum) by using Agrobacterium tumefacien mediated method.The transgenic plants were confirmed that Hf 2 gene was integrated into lily genome by PCR and Southern blot.
