摘    要:利用发根农杆菌R1601感染药用植物三裂叶野葛叶片、茎段、芽、叶柄等外植体,诱导毛状根产生,通过PCR扩增和southern印迹试验,证实发根农杆菌Ri质粒的T-DNA片段已整合进植物的核基因组中,这些毛状根可以在无激素的培养基中快速生长.本试验探索了不同的外植体对诱导率的影响,对产生的毛状根活性物质的含量进行了分析,结果表明,葛转化中叶片是最合适的外植体,而不同的毛状根细胞株中葛根素的含量差异较大,平均含量达到4.21%.

Abstract:Hairy roots appeared after the co-culture of various Pueraria phaseoloides explants and Agrobacterium rhizogenes R1601.It was clearly confirmed that the T-DNA segment was integrated into the cells of hairy roots through PCR amplification and southern blotting.These transformants could grow rapidly on MS medium without hormone.This study also demonstrated the action of explants on the induction rate.The result showed that leaves were superior to other organs as stems,buds and petioles.The puerarin yield of different hairy strain varied largely,holding a 4.21% contents averagely.
