摘    要:将药用野生稻总DNA 通过穗茎注射法导入恢复系R225,创制耐荫、耐光氧化的水稻新种质。结果表明,在T0和T1代幼苗期进行全天侯遮光处理15 d后,有90%~75%的秧苗黄化甚至枯萎;在T2和T3代孕穗期和灌浆结实期,通过测定耐光氧化指数,最终筛选到耐荫、耐光氧化的3 个导入系;经SSR 标记检测,RM219 和RM5490 在导入系中扩增出了与受体R225 不同的3 种带型,表明药用野生稻DNA 被成功导入受体R225 中,并据此推测,标记RM219 和RM5490 很可能与耐荫或耐光氧化QTL 位点紧密连锁。

Study on breeding of new rice germplasm with photooxidationand shade tolerance by using Oryza officinalis
Abstract:Bred new rice germplasm with photooxidation and shade tolerance by ear-stem injecting the DNA of Oryzaofficinalis into the restorer R225. About 90%-75% of the T0 and T1 generation seedlings turned yellow even died after fullyshaded day and night for 15 d. Finally, 3 plant lines with photooxidation and shade tolerance were screened by determiningthe photooxidation-tolerance index in booting and filling stages in T2 and T3 generations. Three types of DNA laddersdifferent from R225 were amplified in transformed lines by SSR makers detecting. The results showed that the DNA of O.officinalis was transformed into R225 successfully. Presumably, RM219 and RM5490 link closely to the QTLs ofphotooxidation or shade tolerance in rice.
