摘    要:为了对半夏种质的鉴定、新品种选育及资源保护提供科学依据,采用形态学性状及ISSR标记,对来自贵州半夏主产区和其他省份的16份野生半夏种质进行了遗传多样性评价。结果表明,筛选出的16条引物共扩增出清晰、重复性好的谱带128条,其中98条为多态性谱带,多态性比率为76.6%;不同样品间的遗传差异性较大,形态学遗传距离变化范围在2.06~9.09,采用UPGMA法可以将16份样品聚为3类;ISSR遗传距离变化范围在0.14~0.53,以0.32为阈值可以将供试样品分为3类。结论:样品的聚类与形态特征密切相关,所选用的形态学性状在半夏资源评价中具有合理性。

Abstract:Based on the polymorphism and reproducibility,16 primers were screened to elucidate the gene diversity of 16 wild P.ternate accessions originated from the main production areas in Guizhou and part of other provinces to provide references for germplasm identification,new variety breeding and resources conservation.The results showed that a total of 128 bands were yielded,among which 98 were polymorphic,and the total percentage of polymorphic loci was 76.6%.Accessions considerably demonstrated high genetic diversity,and the morphological distance between samples ranged from 2.06 to 9.09,and the dendrogram constructed by UPGMA showed that the 16 accessions were classified into three groups.ISSR distance between samples varied from 0.14 to 0.53.Taking 0.32 as a threshold,the tested accessions were also sorted into three clusters based on the ISSR data,and the subcluster of most accessions were highly correlated to their morphological traits,suggesting that the morphological traits used were suitable in the genetic diversity evaluation of the species.
