ACE和ACE2在自发性高血压大鼠肾脏中的表达与作用分析

摘    要:为观察血管紧张素转化酶(ACE)和血管紧张素转化酶2(ACE2)在自发性高血压大鼠肾脏中的表达及它们与高血压形成的关系,取14周雄性Wistar-京都种大鼠(WKY)和自发性高血压大鼠(SHR)各8只,自由采食和饮水1周后测定大鼠收缩压(SBP)、舒张压(DBP)和心率(HR),称重后断头处死,取血液和肾脏组织进行如下试验:放免法测定血浆中血管紧张素Ⅱ(AngⅡ)含量,肾脏组织中ACE、AT1、ACE2和Mas mRNA表达水平的Real-time PCR分析,肾脏组织中ACE2蛋白表达的比较分析。试验结果表明:SHR大鼠SBP和DBP均显著高于WKY大鼠,而心率和体重无显著差异(P>0.05);SHR大鼠血浆中AngⅡ含量和肾脏组织中ACE mRNA表达水平均显著高于WKY大鼠(P<0.05),肾脏组织中ACE2 mRNA和蛋白表达水平均显著低于WKY大鼠(P<0.05),2组大鼠肾脏组织中AT1和Mas mRNA表达没有显著差异(P>0.05)。SHR大鼠肾脏中ACE的mRNA表达显著升高,而ACE2的mRNA和蛋白表达均显著下降。高血压时,肾脏局部组织中ACE和ACE2表达失衡,表现为ACE-AngⅡ-...
