| 摘 要： | 在建立小鼠胚胎干细胞(ES细胞)无饲养层单层培养体系的基础上,使用丁酸钠(NaB)和表皮生长因子(EGF)作为前期诱导分化剂,诱导分化7 d,再用肝细胞生长因子(HGF)和地塞米松(Dex)诱导分化12 d,建立了体外两步法单层诱导ES细胞分化为肝样细胞的方法,并对诱导分化所得的肝样细胞从形态学、基因表达、蛋白分子标记及细胞功能等方面进行了检测,证明了分化所得细胞为肝细胞,肝细胞分化率为41.67%,为肝细胞来源提供了新的方法。
